Tramadol Hydrochloride 6.0 g; Excipients q.s.f 100.00 mL.
Description
Tramadol is a synthetic derivative of codeine, µ agonist, used as an analgesic for a variety of acute and chronic pain syndromes.
Bioactivity
Algen LD has a dual mechanism of action, on one side it joins Mu-opioid receptors and on the other side it inhibits the reuptake of Noradrenaline and Serotonine, thereby increasing the activity of these neurotransmitters. This dual mode of action gives a powerful synergistic action, leading to a comparable analgesia with meperidine or codeine. Its duration of action is around 6-8 hours.
Indications
Algen LD is indicated for moderate to severe somatic and / or visceral pain and for the control of acute or chronic pain (fractures, sprains, contusions, osteoarthritis, neuropathies, etc.).
Algen LD can be administered during anesthetic premedication in order to improve intra-operative analgesia and provide the patient a comfortable awakening.
Algen LD in combination with major tranquilizers increases the sedative action and justifies the dose reduction.
Algen LD is used in the treatment of post surgical pain in outpatients and inpatients, appearing as an effective alternative in most abdominal surgeries, mastectomies, orthopedic and spine surgery.
The combination of Algen LD with nonsteroidal antiinflammatory drugs, increases its effectiveness, producing a multimodal pain relief action.
Dosage
- Dogs and Cats: 1 – 2 mg / kg; SC, IM, Slow IV, Epidural.
- Sport Horses: 1 mg / kg; Slow IV Infusion, Epidural.
Interval between doses: administrate every 6 to 8 hours
Interactions
MAO inhibitors (selegiline) and inhibitors of cytochrome P450 (cimetidine, ketoconazole, erythromycin) can potentiate the toxicity of Tramadol by increasing its plasmatic concentration.
Its administration together with agonists / antagonists (for example buprenorfine, nalbufine) may reduce the analgesic effect.
When combined with α2 agonists, like xylazine or medetomidine, respiratory depression may be observed. In these cases, it is recommended to apply intramuscularly, reduce the final dose of α2 agonists and to consider the need to atropinise the patient.
Contraindications and Limitations of Use
- Prior history of hypersensitivity to the drug.
- Severe hepatic or renal impairment.
- Do not use in pregnant or lactating females.
- Only use in sports horses NOT for human consumption.
- Do not administrate by rapid intravenous infusion or bolus.
- It is contraindicated in the treatment of pain caused by pancreatitis.
- It is contraindicated in patients with uncontrolled epilepsy.
Use with caution in patients who are treated with MAO inhibitors (selegiline) and cytochrome P450 inhibitors (cimetidine, ketoconazole, erythromycin) or who have received it in the course of the last two weeks, because they may potentiate the toxicity of Tramadol by increasing its plasmatic concentration.
Do not administrate together with agonists / antagonists (for example buprenorfine, nalbufine) because it may reduce the analgesic effect.
